MB.02: Danielle Sciorra
Use of FISH to Localize Mouse transgenes implicated in Cardiac Development
Previous researchers created a strain of transgenic mice by inserting exogeneous DNA, which consisited of the Lac Z reporter geneand the transcription factor Ap-2, into mouse embryos. This resulted in an altered phenotype involving cardiac development in the mice. This experiment attempts to determine the nature of, as well as the location of the transgenic mutation in the mouse genome. Through the process of FISH (Fluorescent In Situ Hybirdization), followed by a statistical analysis, the site of the Ap-2 transgene's integration was successfully localized to 13C3.4-D1.5, a non-endogeneous site. The determination that the integration was not endogenous, sustains the hypothized theory that the transgene inserted randomly, and that the site of the transgene's integration is the location of a gene responsible for the phenotypic expression of normal cardiac development. This successful mapping, provides the information needed to clone, and ultimately identify the sequence of genes involved. Through this identification, the mouse sequence can be compared and matched within the human genome, thus finding a novel gene responsible for normal cardiac development in humans.
